Efeito do Bisfenol A a Nível Vascular

Abstract

Este relatório foi elaborado como parte integrante do relatório de estágio do programa curricular do Mestrado Integrado em Ciências Farmacêuticas e é composto por três capítulos distintos: Capítulo I, que diz respeito à investigação, o Capítulo II descreve o meu estágio curricular em farmácia comunitária e o Capítulo III apresenta as atividades desenvolvidas durante o estágio em farmácia hospitalar. O Capítulo I referente à componente de investigação trata o efeito do bisfenol A (BPA) a nível vascular. O BPA é um composto orgânico amplamente utilizado na indústria de plásticos, nomeadamente em recipientes de armazenamento de comida, brinquedos, monómeros odontológicos e equipamentos médicos. Este estudo tem como objetivo analisar os efeitos do BPA nas propriedades contráteis de artérias umbilicais humanas. Recorrendo à técnica de banho de órgãos, anéis de artérias umbilicais humanas, sem endotélio, foram contraídos com serotonina (1µM) e histamina (10µM) e o efeito de diferentes concentrações de BPA (1nM - 100µM) foi analisado. Os resultados obtidos demonstraram que o BPA induz relaxamento da artéria umbilical humana de forma dependente da concentração. Concentrações mais elevadas (50nM, 100nM, 1µM, 10µM e 100µM) provocaram maiores percentagens de relaxamento, enquanto, concentrações mais baixas (1nM e 10nM) induziram um relaxamento menor. Foi também estudado o efeito do BPA na expressão de proteínas envolvidas no processo contráctil, recorrendo ao qPCR. Constatou-se que a incubação com diferentes concentrações de BPA (0,002-20µM) diminuiu a expressão da subunidade alfa dos canais BKCa e do NPRA. Em suma, os resultados obtidos mostram que este disruptor endócrino induz um efeito rápido que consiste na vasodilatação da AUH. Concluiu-se também que o BPA tem efeitos genómicos que podem contribuir para modificar a resposta contráctil da AUH. O Capítulo II está relacionado com o estágio em Farmácia Comunitária realizado em Avenida Farmácia, no Fundão, entre 26 de janeiro e 17 de abril de 2015, sob a orientação da Dra. Anabela Rodrigues. Este capítulo descreve as atividades realizadas durante este período, descrevendo o funcionamento da farmácia comunitária e as tarefas e responsabilidades atribuídas ao farmacêutico. Durante o meu estágio foi-me dada uma grande autonomia na realização de tarefas, que acabou por ser muito relevante para o desenvolvimento de todas as atividades do farmacêutico, essenciais para o meu futuro. O Capítulo III refere-se às atividades desenvolvidas durante o estágio em Farmácia Hospitalar, que teve lugar nos Serviços Farmacêuticos do Centro Hospitalar Cova da Beira, entre 20 de abril e 12 de junho, sob a orientação da Dra. Olímpia Fonseca. Este capítulo descreve as várias áreas onde se integra o farmacêutico hospitalar.

This report was prepared as part of the traineeship of the Pharmaceutical Sciences master’s degree program and consists of three distinct chapters: Chapter I which concerns research, Chapter II describes my professional internship in community pharmacy and Chapter III describes the activities developed during the traineeship in hospital pharmacy. Chapter I refers to the research component, related to the effect of bisphenol A in vascular sistem. BPA is an organic compound widely used in plastics industry, including food container, toys, dental monomers and medical equipment. The present study aims to analyse the effects of BPA on contractile properties in human umbilical arteries. Using standard organ bath techniques, rings of human umbilical arteries without endothelium were contracted by serotonin (1µM) and histamine (10µM) and the effect of different concentrations of BPA (1nM - 100µM) was analyzed. The results showed that BPA induces relaxation a concentration-dependent way. The higher concentrations of BPA (50nM, 100nM, 1µM, 10µM and 100µM) induced higher levels of relaxation and the lower concentrations (1nM and 10nM) induced a smaller relaxartion. The effect of BPA on the expression of different protein involved in the contractile activity was also studied using Real-time PCR. The incubation with BPA (0,002-20µM) decreased the expression of alpha subunit of the BKCa channels and also decreased the expression of NPRA. In summary, the results show that this endocrine disruptor induces a rapid effect consisting in vasorelaxation. Moreover, this study also shows that BPA also have genomic effects that can contribute to modify the contractile vascular response of human umbilical artery. Chapter II is related to the internship in community pharmacy held in Avenida Pharmacy, in Fundão, between January 26th and April 17th of 2015, under the guidance of Dra. Anabela Rodrigues. This chapter describes the activities undertaken during this period, describing the functioning of community pharmacy and the tasks and responsibilities assigned to the pharmacist. During my traineeship I was given a lot of autonomy in performing almost tasks and it was very important for the development of all pharmaceutical skills essential to my future. Chapter III describes the activities developed during the internship in Hospital Pharmacy that took place in the Pharmaceutical Services of Centro Hospitalar Cova da Beira, between April 20th and June 12th, under the guidance of Dra. Olímpia Fonseca. This chapter describes the various areas where it integrates the hospital pharmacist.
