Abstract

Este relatório de estágio encontra-se dividido em dois capítulos: um relativo à componente de investigação laboratorial e outro relativo ao estágio curricular em farmácia comunitária. As infeções provocadas por Candida, designadas por candidoses, são muito prevalentes, afetando cerca de 75% das mulheres durante a idade fértil. Uma das estirpes mais associada a este tipo de infeção é Candida albicans, possuindo muitos fatores de virulência entre os quais a possibilidade de formar biofilmes. Os biofilmes são estruturas multicelulares envolvidas em substâncias poliméricas extracelulares, produzidas pelas próprias células, formando uma matriz. Os biofilmes, por constituírem uma barreira à penetração dos compostos antifúngicos, protegem as leveduras que se encontram inseridas nesta matriz, contribuindo para a virulência de Candida. Desta forma, torna-se essencial descobrir novas terapêuticas eficazes contra este fator de virulência. Este trabalho surge assim como uma possibilidade para o desenvolvimento de uma terapia sinérgica entre o clotrimazol e a tripsina, de forma a possibilitar a destruição de biofilmes. Neste projeto utilizou-se um modelo ex vivo suíno para testar diferentes concentrações de tripsina, clotrimazol e da associação de ambos, em biofilmes preformados de Candida albicans. Procedeu-se também à determinação da atividade enzimática da tripsina sobre a caseína, assim como se testou a viabilidade das células expostas a tripsina e clotrimazol, recorrendo à redução do MTT. O estágio curricular em farmácia comunitária, realizado na farmácia de Bodiosa, em Viseu, decorreu entre 26 de janeiro e 13 de junho, e permitiu-me integrar uma equipa de trabalho focada no utente e no seu acompanhamento. O capítulo II descreve o funcionamento de uma farmácia comunitária e todas as atividades que realizei na mesma: geração e receção de encomendas, armazenamento, atendimento ao público, medição de parâmetros fisiológicos, entre outros.

This report is divided into two chapters: one related to the laboratorial research and the other concerning the internship in community pharmacy. Infections caused by Candida, called candidiasis, are very prevalent, afecting about 75% of women during childbearing age. Candida albicans is the strain more frequently related to this infection, exhibiting many virulence factors including the ability to form biofilms. Biofilms are multicellular structures envolved in an extracellular polymeric matrix. Biofilms are very important to Candida virulence, since they are quite resistant to antifungal therapy. This is due to the biofilm ability of sequester the antifungal agents. So, it becomes mandatory to find out new efective therapies against this virulence factor. This work emerges as a possibilty to the development of a synergistic therapy between clotrimazole and trypsin, in order to enable the destruction of biofilms. In this project, it was used an ex vivo porcine model to test diferent concentrations of trypsin, clotrimazole and the association of both, on preformed Candida albicans biofilms. The determination of the enzimatic activity of trypsin on casein it was also proceeded, as well as it was tested the cell viability exposed to trypsin and clotrimazole, using MTT reduction. The internship on community pharmacy at Bodiosa pharmacy, in Viseu, occurred between january 26th and june 13th and allowed me to integrate a working team focused on patients and their follow-up. The second chapter describes the functioning of a community pharmacy and all the activities that I performed: orders, storage, product dispense, measurement of physiological parameters, among others.
