Abstract

O bisfenol A (BPA) é um composto químico amplamente utilizados em plásticos e resinas e está presente em diversos produtos do dia a dia como embalagens alimentares, brinquedos e utensílios de cozinha. Apesar da sua resistência e durabilidade, este composto é considerado um disruptor endócrino e está associado a diversas patologias, nomeadamente a nível dos sistemas respiratório, imunitário e cardiovascular. Perante a necessidade de substituir e retirar o BPA do mercado, as indústrias recorreram a monómeros estruturalmente semelhantes, nomeadamente o bisfenol F (BPF) e o bisfenol S (BPS), cujos efeitos para a saúde humana são menos conhecidos. Deste modo, o objetivo principal deste trabalho foi estudar os efeitos do BPA, BPF e BPS no tónus arterial e analisar quais os possíveis mecanismos envolvidos nesses efeitos. Os resultados deste trabalho parecem demonstrar que estes compostos exercem efeitos relaxantes independentes de endotélio nas artérias umbilicais humanas (HUA), efeitos que também podem ser observados em células musculares lisas da artéria umbilical humana (HUASMC). A nível genómico verificou-se que tanto o BPA, como o BPF e o BPS, podem interferir com os principais mecanismos responsáveis pelo relaxamento da HUA, através da via NO/sGC/cGMP/PKG e através da modulação da atividade dos canais de Ca2+ do tipo L (LTCC). Assim, a substituição do BPA pelos seus análogos não parece ser benéfica para a saúde cardiovascular humana.

Bisphenol A (BPA) is a chemical compound widely used in plastics and resins and is present in many daily products such as food packaging, toys and kitchen utensils. Despite its resistance and durability, this compound is considered an endocrine disruptor and is associated with various pathologies, particularly in the respiratory, immune and cardiovascular systems. Faced with the need to replace and remove BPA from the market, industries have turned to structurally similar monomers, namely bisphenol F (BPF) and bisphenol S(BPS), whose effects on human health are less well known. The main objective of this study was therefore to study the effects of BPA, BPF and BPS on arterial tone and to analyze the possible mechanisms involved in the effects. The results of this study seem to show that these compounds exert endotheliumindependent relaxing effects on human umbilical arteries (HUA), effects that can also be observed in human umbilical artery smooth muscle cells (HUASMC). At the genomic level, it was found the BPA, BPF and BPS can interfere with the main mechanisms responsible for HUA relaxation, through the NO/sGC/cGMP/PKG pathway and by modulating the activity of L-type Ca2+ channels (LTCC). Thus, replacing BPA with its analogues does not appear to be beneficial for human cardiovascular health.
