Abstract

Neste estudo o conteúdo em alelos S de incompatibilidade de duas cultivares portuguesas de cerejeira com elevado interesse devido às características do seu fruto, a „De Saco da Cova da Beira‟ e „De Saco do Douro‟, foi analisado por processos moleculares a partir de DNA genómico de cada cultivar, utilizando iniciadores específicos na amplificação das sequências codificadoras dos alelos S. Na „De Saco da Cova da Beira‟ foram identificados os alelos S3 e S5, identificando-se na „De Saco do Douro‟ apenas o S9. Paralelamente, estabeleceu-se um método expedito e objectivo para avaliar o vingamento das cerejeiras em condições de campo, incidindo o trabalho experimental sobre a cultivar De Saco do Douro. Procedeu-se à polinização controlada desta cultivar com pólen proveniente das cultivares Bing, Van, Burlat e da própria De Saco do Douro. A viabilidade e o potencial de germinação do pólen das polinizadoras foram avaliados com três corantes (IKI, TTC e X-Gal), e através da análise de germinação. Todos os processos revelaram baixas percentagens de viabilidade do pólen, provavelmente devido à sua imaturidade, que poderá ter resultado das baixas temperaturas ocorridas em Abril. A percentagem de vingamentos obtidos foi baixa, em concordância com os testes efectuados à qualidade do pólen
