Abstract

As metaloproteinases da matriz são um grupo de endopeptídases zinco-dependente responsáveis pela degradação de componentes da matriz extracelular, como o colagénio. Em condições fisiológicas normais atuam em processos de morfogénese e remodelação de tecidos e a sua actividade é regulada a vários níveis, no entanto, quando ocorre um desequilíbrio entre as metaloproteinases e os inibidores podem ser desencadeados inúmeros processos patológicos como, artrite, periodontite, entre outros. A sua acção de degradação do colagénio levou a falhas nas restaurações dentárias uma vez que este é responsável pela ligação resina-dentina. Assim, surgiu a necessidade de incorporar inibidores exógenos das metaloproteinases nas resinas dentárias. Para o desenvolvimento de novos inibidores das metaloproteinases, é necessário analisar pormenorizadamente a sua afinidade com o local de acção e outras propriedades. Estudos efectuados revelaram que determinados monómeros utilizados em resinas são tóxicos, assim, é importante testar a capacidade que um composto tem de atravessar a barreira hematoencefálica e exercer actividade nas células do sistema nervoso central.
