Diagnóstico de fasciolose

Abstract

TÍTULO DO RELATÓRIO: Fasciolosis diagnostic. ABSTRACT The traineeship was held in Coimbra School of Agriculture, part of a project QREN with an enterprise. The aim of the current project was the development of new diagnostic techniques for fasciolosis. This work had the aim to evaluate recombinant proteins homologous of the antigen E/S of Fasciola hepatica for the diagnostic of this parasitosis. An assay was carried out where femal sheep Merino, of 10 months old were infected with 120 metacercarae of F. hepatica. The immunological evaluation was done by ELISA for detection of antibodies anti- Fasciola hepatica using the following antigens: E/S, Cat L Fh8-cys, Fh22 and Prx. Simultaneously, the evaluation of some parasitological and haematological parameters were accomplished. Cat L and E/S showed to have a better performance. At the 5th week p.i., the sensitivity was of 67.5% and specificity of 100%, and for E/S, 87.5% and 100 % respectively. At 11th weeks p.i., the sensitivity and specificity were of 85.7% and 100% for the first and 100% sensitivity and specificity for the second. The elimination of eggs in the feaces started between the 9th and 11th week p.i., and others parasites were present, such as Eimeria sp and gastro-intestinal strongyles which did not affect the quality of serological diagnosis. Keywords: ELISA, antibodies, proteins, parasites, specificity, sensitivity.
