A aplicação de células estaminais na medicina regenerativa

Abstract

As células estaminais pluripotentes, isto é, as células estaminais embrionárias e as células estaminais pluripotentes induzidas (iPSCs) têm como propriedades principais e distintivas as capacidades de se autorrenovarem e diferenciarem em diferentes tipos celulares. Ao longo do tempo as células estaminais embrionárias revelaram possuir as qualidades exigidas para fins de investigação científica e para a área da medicina, no entanto, ao contrário das iPSCs, suscitam sérias questões éticas que limitam a sua aplicação na prática clínica. A descoberta das iPSCs trouxe a possibilidade de converter células diferenciadas em células estaminais multipotentes com a capacidade de dar origem a todos os tipos celulares através de técnicas de reprogramação celular. Existem variadas aplicações onde estas células podem incidir, incluindo nos modelos de doença in vitro para o estudo dos mecanismos de doenças, para a pesquisa e descoberta de fármacos e na medicina regenerativa. As iPSCs constituem uma grande promessa na medicina regenerativa, sobretudo ao nível da terapia celular de substituição. No entanto ainda não são totalmente compreendidos os mecanismos subjacentes a esta aplicação e ainda existem bastantes riscos associados que ainda não são suficientemente controlados para evitar reações indesejadas. Neste trabalho é feita uma revisão sobre as áreas de aplicação desta tecnologia, incidindo sobretudo na medicina regenerativa e no estado atual da sua aplicação na prática clínica, bem como os aspetos que existem a melhorar para que esta constitua uma terapia celular segura e eficaz num futuro próximo.
