A influência da serotonina na fisiologia da depressão

Abstract

A depressão é um distúrbio mental que afeta cerca de 350 milhões de pessoas em todo o mundo. É estimado que em 2030 será a maior causa de incapacidade, o que faz desta um notório problema de saúde pública. É uma doença multifatorial, sendo que a sua fisiopatologia permanece pouco esclarecida. De todas as hipóteses consideradas, a deficiência de monoaminas e alterações morfológicas de estruturas cerebrais são as mais relevantes. O sistema serotoninérgico é o principal alvo dos tratamentos farmacológicos atuais. Pesquisas recentes demonstraram que a serotonina (5-HT) não atua diretamente no humor, mas sim no enviesamento negativo do processamento de informação emocional, que predispõe o indivíduo a sofrer de depressão. Foi descoberta ainda uma associação entre a inflamação periférica e a indução da enzima que metaboliza o precursor da 5-HT, o envolvimento da amígdala e do microbiota na depressão. A fim de contornar a demora na resposta e falha no tratamento, há necessidade de novas descobertas de métodos de tratamento e de marcadores de vulnerabilidade à doença (como o antigénio melanoma gene-D1 e o polimorfismo no gene SLC6A4). O farmacêutico e demais profissionais de saúde têm um papel essencial na educação do doente quanto à sua depressão, estimulando-o para estratégias que contribuam para uma recuperação mais rápida e eventual prevenção de episódios futuros.
