Caracterização do microbioma humano

Abstract

O ser humano é constituído por dez vezes mais bactérias que células humanas. Este deixa de ser estéril a partir do momento em que nasce, sendo colonizado por bactérias dependendo do tipo de parto e da dieta. Existe uma grande diversidade microbiana intra e interpessoal, apesar destes microrganismos poderem sofrer alterações devidas a diversos fatores, conduzindo a doenças. No entanto, os microrganismos comensais desempenham funções importantes e beneficiam de certa forma o seu hospedeiro. O Projeto Microbioma Humano (HMP) tem como objetivo a caracterização e identificação dos microrganismos que habitam o ser humano e o estudo de fatores que provocam alterações destes. Para tal, têm surgido novas técnicas de estudo do microbioma humano de forma a ultrapassar as limitações de cultivo de certas espécies bacterianas. Nesta dissertação foi analisada a diversidade do microbiota na cavidade oral, pele, trato urogenital e trato gastrointestinal. No entanto, o trato gastrointestinal é o que apresenta uma maior diversidade. Esta grande diversidade tem melhorado o perfil metabólico e respostas imunológicas. Além disso, o microbiota tem demonstrado viabilidade de auxílio na terapêutica de doenças através da estimulação do sistema imunitário. Por outro lado, quando o microbioma sofre alterações, este fica mais susceptível a certas infeções. No caso do microbioma vaginal, quando este sofre desequilíbrios pode desencadear vaginose bacteriana, ficando a mulher mais susceptível a infeção por VIH quando em contato com o vírus. Nesta dissertação verifica-se que os microrganismos não são sempre patogénicos como a maioria das pessoas pensam, podendo virem a ser a solução para várias doenças.
