Bioterapia da infeção por SARS-CoV-2 com nanobodies geneticamente modificados

Abstract

Apesar de o combate à COVID-19 ser atualmente apenas preventivo (vacinação) existe uma necessidade urgente de novas soluções terapêuticas. Este trabalho, propõe-se a contribuir para o desenvolvimento de estratégias terapêuticas inovadoras através da utilização de nanobodies e SPIKES recombinantes (rNano e rSPIKE), entre outros. A metodologia utilizada baseou-se na produção de rNano e rSPIKE produzidos por engenharia genética. Posteriormente, procedeu-se à purificação destas proteínas recombinantes realizando uma série de testes para avaliar a sua eficácia neutralizadora no SARS-CoV-2. Esta avaliação foi realizada em culturas de células de pulmão (A549) e em macrófagos (RAW264.7). Assim, foi avaliado o efeito inibitório de diferentes tratamentos: ramipril (inibidor farmacológico da ACE2) rSPIKE (inibidor competitivo do SARS-CoV-2); rNano (neutralizador do SARS-CoV-2). Foram ainda avaliadas interleucinas envolvidas na cytokine storm e determinantes do stress oxidativo. O estudo revelou que, de todos os potenciais inibidores do SARS-CoV-2 o que apresentou maior efeito terapêutico foi i rNano com uma taxa de redução viral de 44% majorada para 52% em combinação com o ramipril. A IL-1β parece ser um bom biomarcador da neutralização terapêutica deste vírus e as glutationas parecem ser bons biomarcadores da evolução da doença. Este trabalho é uma contribuição humilde para o desenvolvimento de terapias anti-COVID-19.
