Abstract

Objetivos: Testar um protocolo clínico de recolha de dados e de amostras biológicas para analisar o microbioma oral de utentes da CDU-UCP com questões específicas para portadores de prótese removível. Materiais e métodos: O estudo incluiu 24 pacientes sendo 12 portadores de prótese removível e 12 não portadores. Foram recolhidas amostras de saliva, biofilme dentário, esfregaços das próteses removíveis e da mucosa oral em contato com as mesmas. As análises do ADN microbiano extraído das amostras foram feitas por PCR quantitativo e analisadas quanto à carga total bacteriana. Resultados: Os pacientes observados apresentavam uma heterogeneidade de reabilitações incluindo próteses totais e parciais, esqueléticas e acrílicas. A quantidade total de carga bacteriana na saliva não apresentou diferenças estatisticamente significativas entre os pacientes com e sem prótese removível. No entanto, a carga bacteriana média na saliva é superior nos pacientes com prótese removível. A carga total bacteriana quantificada na saliva foi sempre superior à encontrada, no mesmo individuo, através da zaragatoa. Conclusão: Trabalhos aliando os aspetos moleculares às descrições clínicas de saúde oral são possíveis de realizar na FMD-UCP, mas é necessário melhorar o recrutamento de pacientes para participação nos estudos. No que diz respeito à recolha de biofilme oral das próteses para análise do microbioma é recomendado alterar alguns procedimentos para aumentar a concentração de microrganismos recolhidos.

Aims: To test a clinical protocol for data and biological samples collection for the analysis of the oral microbiome of patients at the CDU-UCP. Specific questions and sample collection protocols are tested or patients with removable dentures. Materials and methods: The study included 24 patients, 12 of which were removable denture wearers and 12 non-users. Samples of saliva, dental biofilm, smears from removable dentures and oral mucosa in contact with them were collected. The analyzes of the microbial DNA extracted from the samples was performed using quantitative PCR and analyzed for the total bacterial load. Results: The patients observed had heterogenous rehabilitations including complete and partial, metal and acrylic dentures. The total amount of bacterial load in saliva showed no statistically significant differences between patients with and without dentures. However, the average bacterial load was higher in patients with dentures. The total microbial load quantified in saliva was always higher than that found in the same individual using the swab. Conclusion: Investigations combining molecular and clinical aspects of oral health are feasible at the FMD-UCP, but it is necessary to improve the recruitment of patients to participate in the studies. Regarding the collection of oral biofilm from dentures for microbiome analysis, it is recommended to change some procedures to increase the concentration of collected microorganisms.
