O proteoma do microbioma oral humano : que funções exercem as proteínas microbianas?

Abstract

A base de dados OralOme, consultável na interface OralCard, reúne toda a informação atualizada, resultante dos estudos de proteómica de tecidos orais e estudos in vitro. Além de proteínas do hospedeiro inclui proteínas produzidas pelo microbioma oral sendo possível conhecer as proteínas produzidas por cada género e/ou espécie. Esta ferramenta bioinformática pretende ajudar no desenvolvimento de novos métodos diagnósticos, prognósticos ou na pesquisa de agentes terapêuticos. Objetivo: De acordo com os dados de proteómica de tecidos orais e estudos in vitro depositados no OralCard, foi realizada uma análise crítica do papel funcional desempenhado pelas proteínas de origem bacteriana dando mais ênfase áquelas envolvidas no processo biológico da patogénese. Materiais e métodos: As proteínas do microbioma oral foram analisadas quanto aos processos biológicos em que participam, sendo feita a análise funcional das proteínas classificadas com o GO “pathogenesis”. Resultados: Nas 10428 proteínas bacterianas analisadas, 298 foram classificadas com o GO “pathogenesis”. Destas, apenas 44 estão caracterizadas a nível da proteína sendo que as restantes têm evidência a nível do gene. A análise revela que destas 298 apenas 11 foram identificadas em amostras da cavidade oral sendo as restantes identificadas in vitro. Os mecanismos específicos de virulência com mais número de proteínas associadas são a sobrevivência do microrganismo dentro da célula do hospedeiro e a lise da célula hospedeira. Conclusão: Os mecanismos específicos da patogénese para a maioria das proteínas do microbioma oral encontram-se bem relatados. No entanto, quase todas as proteínas foram descritas em estudos in vitro e não em amostras da cavidade oral. Seria assim pertinente em trabalhos futuros, comprovar a existência destas proteínas em amostras salivares de pacientes com as patologias orais para as quais os microrganismos responsáveis pela sua produção estão envolvidos.

In the OralOme database, available through the OralCard interface, information resulting from proteomics studies of oral tissues and in vitro studies is gathered and updated. Besides the host proteins, proteins produced by the oral microbiome are includes ant it is possible to know which are produced by each genre and / or species. This bioinformatics tool facilitates the development of new diagnostics, prognostics and the search for new therapeutic agents. Objective: Using the data deposited in the OralCard relative to the proteins produced by the oral microbiome, a critical analysis of the functional role performed by the proteins of bacterial origin was conducted. Emphasis was given to proteins involved in the pathogenesis biological process. Materials and methods: The oral microbiome proteins were analyzed as to the biological processes in which they participate. A functional analysis of the proteins classified with the GO "pathogenesis" was performed using public databases. Results: Of the 10 428 bacterial proteins analyzed, 298 were classified with the GO "pathogenesis". Of these, only 44 are characterized at the protein level and the remaining, have evidence at the gene level. The analysis revealed that only 11 of the 298 were identified in samples from the oral cavity and the remaining identified in vitro. The specific mechanisms of virulence with more number of associated proteins are the survival of the organism within the host cell and it’s lysis. Conclusion: The specific mechanisms of the pathogenesis for most oral microbiome proteins are well described. However, almost all proteins have been described in vitro studies. Therefore, it would be desirable that future studies, reveal the existence of these proteins in saliva samples from patients.
