Abstract

A carboxilase dependente da vitamina K (VKD), permite que ocorra uma transformação dos resíduos de ácido glutamico (Glu) para ácido carboxiglutamico (Gla), que torna funcionais as proteínas dependentes da vitamina K (VKD), influenciando a fisiologia, a homeostasia, a apoptose, a transducção do sinal, o controlo do crescimento, a calcificação arterial e o desenvolvimento do esquelto. A proteina Gla da matriz (Mgp) e a osteocalcina (Oc ou proteína Gla do osso- Bgp), têm a capacidade de ligar o cálcio através do dominio contendo os resíduos gla, sendo uma função essencial para a sua actividade biológica como inibidor de calcificação (Mgp) e modulador dos cristais de hidroxiapatite (Oc). Usaram-se fases larvares de peixe zebra (Danio rerío), para compreender o efeito da varfarina no desenvolvimento esquelético. A varfarina é um conhecido anticoagulante, que inibe a acção da carboxilação da vitamina K (VKGC), inibindo a maturação da Oc e da Mgp e levando a um incorrecto resultado da sua função. Para avaliar os efeitos varfarina, recorreu-se à dupla coloração do esqueleto com azul de alcian e vermelho de alizarin, revelando uma inibição da mineralização. A imunodetecção da Oc e da Mgp revelaram uma acumulação acrescida em tecidos calcificados em desenvolvimento. A quantificação da expressão por PCR em tempo real revelou uma sobre-expressão de mgp e oc por resposta ao tratamento com varfarina. A varfarina aparenta afectar a correcta calcificação das larvas e na função da Oc e Mgp.
