Abstract

En este trabajo se describe la obtención y caracterización de una serie de anticuerpos monoclonales frente a distintos marcadores de diferenciación leucocitarios porcinos, entre los que cabe destacar CD1 1a, CD18, CD29 y CD5. Para su obtención se siguieron dos estrategias de inmunización, utilizando como inmunógenos tanto leucocitos porcinos completos como una proteína recombinante (CD29R), mientras que para su caractización se recurrió a técnicas como el ELISA, citometría de flujo, inmunoblotting, inmunoprecipitación e inmunohistoquímica. Asimismo, se discuten las posibles aplicaciones que podrían tener estos anticuerpos en diversos estudios relacionados con el xenotrasplante, la Sanidad Animal y la fisiología de los leucocitos porcinos. También se caracterizaron parcialmente otros anticuerpos frente a antígenos linfocitarios y plaquetarios porcinos. Por otra parte, se describe por primera vez la reacción de un anticuerpo monoclonal frente a PRPC murino con PrPC porcino y se emplea en el estudio, mediante citometría de flujo, de la expresión de PrPC en lecuocitos y plaquetas de las especie porcina, señalando su peculiaridades con respecto al patrón de expresión en otras especies animales. Se discuten las posibles aplicaciones prácticas de dicho anticuerpo en estudios sobre encefalopatías espongiformes humanas en que se empleara al cerdo como modelo animal.
