Estudio multigeneracional de los efectos del Bisfenol A como disruptor endocrino

Abstract

El Bisfenol A es un contaminante emergente de gran ubicuidad en el medio ambiente al que los seres vivos están expuestos de forma continuada e inadvertida. Está considerado como una sustancia con capacidad disruptora endocrina que provoca alteraciones en el sistema reproductivo, inmunológico y neurológico entre otros, así como alteraciones metabólicas sobre los organismos expuestos a él. Nuestro estudio tuvo como objetivos, por una parte y mediante una exposición a diferentes dosis de BPA en sucesivas generaciones de ratones, evaluar los efectos de este disruptor endocrino sobre el desarrollo de las crías (F1, F2, F3 y F4), mediante el análisis de diferentes parámetros en las camadas, así como determinar en los parentales las posibles alteraciones sobre distintos parámetros bioquímicos sanguíneos tras su exposición al BPA para relacionarlo con los posibles efectos tóxicos y evaluar los efectos histopatológicos a nivel hipofisario y gonadal. Por otra parte, mediante una exposición a largo plazo y bajas dosis de BPA en ratones adultos, evaluar las posibles alteraciones crónicas en los niveles de distintos parámetros bioquímicos sanguíneos para relacionarlos con los posibles efectos tóxicos derivados de esta exposición al BPA. Los resultados mostraron que, a lo largo de diferentes generaciones, no hubo modificaciones en los parámetros reproductivos evaluados, como la duración del ciclo estral, el tamaño de las camadas o el porcentaje de crías vivas al destete. En consonancia con estos resultados, no se evidenciaron alteraciones histopatológicas en los órganos analizados. Sin embargo, sí se demostró que existían alteraciones en parámetros bioquímicos como la glucosa, cuyos niveles aumentaron en las diferentes generaciones de reproductores a las mayores dosis de exposición, así como en la F4 descendiente; proteínas totales, que disminuyeron a las dosis más altas en la F1 y F3 reproductora y F3 descendiente, y albúmina, que experimentó un incremento en los animales expuestos a 50 μg/kg/d en todas las generaciones de reproductores y en la progenie, por lo que estos parámetros podrían considerarse como indicadores indirectos del efecto temprano sobre la salud provocado por este compuesto que además se transmite a través de sucesivas generaciones. Los resultados obtenidos en el estudio de exposición a largo plazo mostraron que existieron alteraciones en diferentes parámetros bioquímicos sanguíneos que inicialmente podrían indicar una posible alteración a nivel hepático y renal de los animales expuestos de forma crónica a este disruptor endocrino.

Bisphenol A (BPA) is an emerging contaminant of great ubiquity in the environment to which living organisms are continuously and inadvertently exposed. It is considered a substance with endocrine-disrupting capabilities, causing alterations in the reproductive, immune, and neurological systems, among others, as well as metabolic disturbances in organisms exposed to it. Our study aimed, on one hand, through exposure to different doses of BPA across successive generations of mice, to evaluate the effects of this endocrine disruptor on the development of offspring (F1, F2, F3, and F4) by analyzing various parameters in the litters, as well as to determine potential alterations in various blood biochemical parameters in the parental generation after BPA exposure, to relate these to possible toxic effects and evaluate histopathological effects at the pituitary and gonadal levels. On the other hand, through longterm exposure to low doses of BPA in adult mice, the study aimed to assess possible chronic alterations in the levels of various blood biochemical parameters to relate these to potential toxic effects arising from BPA exposure. The results showed that, across different generations, there were no modifications in the reproductive parameters evaluated, such as estrous cycle duration, litter size, or the percentage of live pups at weaning. Consistent with these results, no histopathological alterations were evident in the analyzed organs. However, it was demonstrated that there were alterations in biochemical parameters such as glucose, whose levels increased in the different generations of breeders at higher exposure doses, as well as in the F4 offspring; total proteins, which decreased at the highest doses in the F1 and F3 breeders and F3 offspring; and albumin, which increased in animals exposed to 50 μg/kg/day in all generations of breeders and their progeny. Therefore, these parameters could be considered as indirect indicators of early health effects caused by this compound that is also transmitted across successive generations. The results obtained from the long-term exposure study showed alterations in various blood biochemical parameters that could initially indicate a possible liver and renal disorder in animals chronically exposed to this endocrine disruptor.
