En este trabajo se estudi? el efecto de la incorporaci?n en la dieta de una mezcla de aceites rica en ?cidos grasos ?-3, ?-6 y ?-9 (Ometrix?) sobre los cambios que se generan por la hipertensi?n y/o la hipercolesterolemia en ratas.Ometrix? se incorpor?, al 10%, a una dieta hipercolesterol?mica que recibieron ratas espont?neamente hipertensas (SHR) y normotensas durante 2 meses. Otro estudio se realiz? en ratas SHR que recibieron durante 1 mes la dieta hipercolesterol?mica y posteriormente se incorpor? a ?sta durante 2 meses m?s la mezcla de aceites.El tratamiento redujo la presi?n arterial de ratas SHR sin modificarla en normotensas. Previno o redujo el incremento de los niveles plasm?ticos de colesterol total y LDLc y la disminuci?n de los niveles de HDLc provocados por la dieta hipercolesterol?mica en ratas hipertensas y normotensas. En normotensas adem?s se redujeron los triglic?ridos. En ambos casos mejor? el ?ndice de riesgo cardiovascular. Se observ? una recuperaci?n de la disfunci?n endotelial provocada por la hipertensi?n y/o la hipercolesterolemia al incrementar la relajaci?n dependiente de endotelio. En este efecto podr?a estar implicada la producci?n de prostanoides por la v?a de la ciclooxigenasa. Redujo la hipertrofia cardiaca y vascular en ratas SHR. En arterias de conductancia principalmente redujo el grosor de la pared y en arterias de resistencia aument? el lumen. El tratamiento mejor? el estado redox en ratas hipertensas e hipercolesterol?micas incrementando los niveles de glutati?n total y disminuyendo los de glutati?n oxidado en h?gado, adem?s se observ? un incremento en la actividad enzim?tica de super?xido dismutasa, catalasa y glutati?n reductasa.En ratas con da?o hep?tico agudo inducido por tec-butilhidroper?xido, Ometrix? impidi? la elevaci?n de la actividad de enzimas plasm?ticas indicadoras de este da?o, disminuy? el da?o a nivel histol?gico en h?gado y los cambios en la actividad de la catalasa y la glutati?n reductasa.
We studied the effect of a ?-3, ?-6 and ?-9 fatty acids mixture (Ometrix?) on the alterations observed in rats with hypertension and/or hypercholesterolemia. Ometrix? was incorporated, in a 10%, on a hypercholesterolemic diet which was used to feed SHR and normotensive rats during 2 months. In other study, SHR were fed for the first month with the hypercholesterolemic diet and then Ometrix? was incorporated into hypercholesterolemic diet for another two months.Treatment reduced systolic blood pressure in SHR without changes in normotensive rats. It prevented or reduced the increase of total cholesterol and LDLc in plasma, and the decrease of the HDLc that the hypercholesterolemic diet caused in hypertensive and normotensive rats. In normotensive rats it also reduced triglycerides levels. The treatment improved the atherogenic index in both cases. The impaired endothelial function by hypertension and/or hypercholesterolemia was also improved, that effect might be related with prostanoids production. Ometrix? reduced the cardiac and vascular hypertrophy in SHR. In aorta, it reduced the medial layer thickness and in resistance arteries increased the lumen. Treatment improved the hepatic antioxidant status in hypertensive and hypercholesterolemic rats. It increased total glutathione and decreased disulfide glutathione (GSSG). An increase on the enzymatic activity of superoxide dismutase, catalase, and glutathione reductase was also observed.In rats with tert-butylhydroperoxide-induced hepatic damage, Ometrix? prevented the increase of alanine aminotransferase, aspartate aminotransferase and lactate dehydrogenase enzymatic activity, improved histological damage in liver and prevented the changes on catalase and glutathione reductase.
