Desarrollo de análogos del ácido litocólico modificados en la cadena lateral y anillo-a como ligandos activos del receptor de la vitamina D

Abstract

Se han diseñado por docking y sintetizado los compuestos CARR y CAR-S que se comportan como potentes agonistas del ácido litocólico de potencial interés terapéutico para el tratamiento de enfermedades hiperproliferativas. Los análogos CAR-R y CAR-S se sintetizaron en 13 pasos a partir del ácido litocólico y rendimientos globales del 17% y 18%, respectivamente. Posteriormente, se han estudiado tres estrategias de síntesis distintas para acceder a cadenas laterales ciclohexánicas dihidroxiladas de análogos modificados en el anillo A del ácido litocólico. La ruta sintética que se basa en la transposición de diaceno conduce con éxito a las cadenas propuestas. Posteriormente, se han descrito dos rutas sintéticas del diol 31a, precursor de los análogos GLD y GLS diseñados por docking, a partir del ácido litocólico.
