Abstract

As paredes celulares primarias de plantas conforman unha estrutura moi dinámica e versátil implicada na regulación do crecemento. Están compostas principalmente por celulosa, pectinas e hemicelulosas como o xiloglucano (XiG). Analizar os efectos da alteración ou eliminación experimental dun ou varios destes compoñentes, permite achegarnos á comprensión da estrutura, dinámica e función da parede primaria. É obxecto deste estudo investigar as respostas de Arabidopsis thaliana fronte ás alteracións da parede primaria, producidas mediante mutantes sen XiG e liñas de sobreexpresión inducible por estradiol. Constatáronse diferenzas significativas claras no crecemento de hipocótilos etiolados, entre mutantes sen XiG e liñas complementadas, o que indica posibles efectos do XiG sobre a extensibilidade (aínda que os resultados non descartan que poida deberse a outros factores). Ademais observouse que o debilitamento moderado da rede de pectinas leva a un crecemento de hipocótilos etiolados significativamente maior respecto ao silvestre. Resultados pouco congruentes suxeriron tamén que a disrupción combinada de XiG e pectinas podería aumentar o crecemento de hipocótilos por un debilitamento maior da parede, o que indicaría a existencia dun posible efecto compensatorio entre as redes de pectina e XiG. Por outra banda, foi implementado con éxito un protocolo de extracción e cuantificación de XiG.
