Influencia de la adenosina y de los receptores de adenosina en la diferenciación de subpoblaciones de monocitos

Abstract

La adenosina (Ado) es una molécula endógena moduladora de la respuesta inflamatoria. Este nucleósido se encuentra a bajas concentraciones en condiciones normales, pero condiciones como la hipoxia, necrosis, estrés metabólico y daño tisular (ej., cáncer u otras enfermedades) incrementan su síntesis y liberación. Ado interfiere en la activación de los macrófagos y en la diferenciación de monocitos a través de cuatro receptores asociados a proteínas G: A1, A2A, A2B y A3. El objetivo de este trabajo fue comprobar la acción que ejerce la adenosina sobre la diferenciación de monocitos a células dendríticas (DCs, Dendritic Cells) así como describir la acción de sus receptores A2A y A2B en dicho proceso. Para lograrlo, se cultivaron poblaciones de monocitos obtenidos a partir de concentrados leucocitarios (BC, Buffy Coats) de donantes sanos en presencia de factor estimulante de colonias de granulocitos y macrófagos (GM-CSF, Granulocyte Macrophague Colony-Stimulating Factor) e interleucina-4 (IL-4, Interleukin-4), un análogo no selectivo y más potente de adenosina, la 5’-(N-Etilcarboxiamido)adenosina (NECA, 5’-(N-ethylcarboxiamido) adenosine) y dos antagonistas de sus receptores: CV69 (A2AR antagonista) y SYAF030 (A2BR antagonista). Los resultados obtenidos indican que la adenosina interfiere en la diferenciación de los monocitos hacia DCs inflamatorias dando lugar a una población de células con características intermedias entre ambos tipos celulares. Sin embargo, no se ha logrado discernir el receptor de adenosina involucrado entre los dos candidatos más probables: A2AR y A2BR
