Efecto de diferentes AINEs sobre la funcionalidad de los condrocitos humanos en cultivo

Abstract

Los mecanismos exactos de la activación de los condrocitos hacia procesos artrósicos así como los mecanismos genéticos involucrados todavía no se comprenden del todo. La artrosis abarca una serie de procesos destructivos de los componentes de la matriz extracelular del cartílago junto con una excesiva producción de citoquinas y enzimas proteolíticas, causando dolor, hinchazón, rigidez y pérdida de la función de la articulación afectada. Entre las diversas agresiones a la articulación que pueden ocasionar la destrucción del cartílago se incluyen la edad, el estrés mecánico, daños previos, y factores que modulan la inmunidad. Como el proceso está causado principalmente por mediadores inflamatorios, los fármacos antiinflamatorios no esteroideos (AINEs) proveen alivio sintomático bloqueando la síntesis de prostaglandinas; es por ello que son ampliamente utilizados a nivel mundial. Se ha analizado el efecto de diferentes AINEs sobre condrocitos humanos sanos y artrósicos en diferentes procesos celulares, componentes de la matriz extracelular, y mediadores inflamatorios involucrados en la degeneración artrósica.
