Abstract

Este capítulo tem como objetivo principal apresentar algumas das técnicas mais importantes usadas em Imunologia para isolar e estudar os diferentes tipos celulares que constituem o sistema imunológico, nomeadamente linfócitos, monócitos e células dendríticas, assim como os mediadores biológicos que produzem. Não pretende ser um manual de laboratório, mas antes um guia que ajude à compreensão de alguma das principais técnicas imunológicas utilizadas em investigação e diagnóstico. O capítulo incidirá, sobretudo, em técnicas imunológicas mais nas áreas da bioquímica e da biofísica (por exemplo, imunoprecipitação de proteínas, microscopia de fluorescência, e citometria de fluxo), referindo mais sucintamente técnicas imunológicas clássicas de deteção e quantificação de antigénios (por exemplo, imunoensaios). Todavia, em primeiro lugar serão abordadas as técnicas de isolamento e cultura de populações de células mononucleares do sangue.
