Abstract

Os linfócitos B expressam à sua superfície um recetor cuja parte responsável pelo reconhecimento do antigénio é uma imunoglobulina. As imunoglobulinas pertencem à superfamília das imunoglobulinas pelo facto de serem constituídas por domínios V e C, com propriedades características. Estas moléculas apresentam uma região variável que confere especificidade para o antigénio e uma região constante com propriedades efetoras. Os genes que codificam para as imunoglobulinas são constituídos após rearranjos somáticos do ADN da linha germinal. Durante a recombinação somática e por mutação somática, mecanismos especializados são responsáveis pela geração de diversidade que explica a existência de um elevadíssimo número de imunoglobulinas com distintas especificidades para o antigénio. Em resposta ao antigénio, os linfócitos B podem efetuar uma mudança no isotipo da imunoglobulina que expressam, podendo assim alterar as propriedades efetoras destas moléculas mantendo a mesma especificidade para o antigénio. A capacidade dos linfócitos B expressarem uma forma membranar ou segregada da mesma imunoglobulina, assim como de expressarem simultaneamente IgM e IgD, deve-se a um processo de splicing alternativo do ARN.
