Abstract

SOMMARIO DELL'INVENZIONE L'invenzione della presente descrizione si basa sulla scoperta che la chemioresistenza in numerose forme leucemiche è eliminata o fortemente ridotta da un trattamento con almeno un composto per terapie antitumorali in combinazione con almeno un bloccante del canale di potassio hERG1. Questi esercitano, con un inatteso effetto sinergico un effetto proapoptotico sulle cellule leucemiche chemioresistenti. La presente invenzione si riferisce quindi a: -nuove combinazioni di uno o piu bloccanti di canali hERG1 e di uno o piu composti per terapie antitumorali, per uso nel trattamento di leucemie chemioresistenti e/o di leucemie potenzialmente chemioresistenti. -composizioni farmaceutiche comprendenti almeno un composto per terapie antitumorali in combinazione con almeno un inibitore del canale di potassio hERG1, per uso nel trattamento di leucemie chemioresistenti e/o di leucemie potenzialmente chemioresistenti; e -metodi terapeutici per il trattamento di dette leucemie con dette composizioni. La presente invenzione si riferisce, inoltre, -al metodo in vitro di co-coltura di cellule leucemiche con cellule stromali midollari comprendente il passaggio di mettere in coltura cellule stromali midollari e aggiungere successivamente cellule leucemiche di linee cellulari o cellule leucemiche primarie. -dette Co-colture cellulari: e -un metodo in vitro di screening per l'identificazione di sostanze o combinazioni di sostanze efficaci per il trattamento delle leucemie chemioresistenti e/o potenzialmente chemioresistenti comprendente i seguenti passaggi: • a) trattare dette co-colture cellulari come precedentemente definite in cui dette cellule leucemiche sono chemioresistenti con una o piu sostanze o combinazioni di sostanze; b) valutare I'apoptosi delle cellule leucemiche dopo il trattamento; c) identificare le sostanze o le combinazioni di sostanze efficaci nel ripristino delle vie apoptotiche.
