Sobreexpresión de citocinas TH17 en la fibrosis pulmonar inducida por administración endotraqueal de adriamicina

Abstract

La bleomicina es un antibiótico genotóxico con actividad antitumoral, que induce fibrosis pulmonar cuando se administra por vía i.v., endotraqueal e incluso subcutánea. Por ello es el modelo experimental de esclerodermia más utilizado. La administración subcutánea de adriamicina o doxorrubicina, otro antibiótico empleado en el tratamiento de múltiples tumores, también provoca fibrosis cutánea en el sitio de punción, pero no afecta al pulmón. No existiendo reportes previos, hemos puesto a punto un modelo de fibrosis pulmonar mediante inyección endotraqueal de adriamicina y hemos analizado los mecanismos inmunológicos que intervienen en el desarrollo de las lesiones. Los resultados de expresión (Q-PCR) de distintos tipos de colágenos, de citocinas y de factores de transcripción específicos de linaje de diferenciación de células T-CD4, muestran un marcado aumento en la expresión de colágeno-I (fibrilar) y colágeno IV (membranas basales) en relación con el incremento en la expresión de citocinas asociadas a células TH17 y del factor de transcripción ROR¿T, específico de este linaje. En las células obtenidas mediante lavado broncoalveolar en estos animales hemos constatado un incremento en la frecuencia de linfocitos CD4+ activados productores de IL-17. Estos resultados sugieren un papel destacado de los linfocitos TH17 en la producción de fibrosis pulmonar por adriamicina.
