Estudi en mostres humanes i en cultius neuronals d’indicadors del sistema glutamatèrgic i dopaminèrgic en relació a l’exposició prolongada a contaminants orgànics tòxics persistents

Abstract

En esta tesis se investigan los efectos que ejercen ciertos contaminantes orgánico tóxico persistentes en indicadores del sistema glutamatérgico y dopaminérgico. Concretamente se estudia 1) el efecto de contaminantes organoclorados y el metilmercurio en el transporte de glutamato en placenta humana y en los niveles de aminoácidos excitadores, glutamato y aspartato, en sangre de cordón umbilical humana, 2) el efecto de la exposición prolongada (>96h) del PCB 138 y el hexaclorociclohexano (HCH) (gamma o beta) en receptores y transportadores glutamatérgicos en cultivos primarios de neuronas corticales de ratón durante el periodo de maduración, y 3) el efecto de la exposición prolongada (>96h) del PCB 138 y el lindano (gamma-HCH) en receptores y transportadores dopaminérgicos en cultivos de neuroblastoma humano SH-SY5Y durante el periodo de diferenciación. Esta tesi demuestra que dichos contaminantes afectan a la normal funcionamento de distintos parámetros del sistema gluatamatérgico y dopaminérgico en distintas muetsras de estudio, cuando se encuentra a dosis subtóxicas y largos periodos de tiempo, y que dichas alteraciones podrian implicar disfunciones en el neurodesarrollo fetal.

Aquesta tesi ha estat possible gràcies a una beca predoctoral IDIBAPS otorgada de gener del 2008 a desembre 2011. Aquest treball ha estat finançat pels següents projectes: “Estudio de mecanismos de neurotoxicidad crónica de contaminantes tóxicos: papel de los receptores GABA-A, de glutamato y de estrógenos. Posibles intervenciones terapéuticas” (PI10/00453); “Identificación de dianas moleculares y funcionales alteradas en procesos de neurotoxicidad inducida por contaminantes ambientales y biocidas en cultivos neuronales” (PI 06/1212); “Determinación de niveles de glutamato y dopamina y de sus transportadores plasmáticos en muestras de sangre y de placenta. Búsqueda de posibles biomarcadores de exposición y efecto para el metilmercurio y otros contaminantes ambientales.” (AA07_012); “Proyecto CIBER Epidemiología y Salud Pública. Neurotoxicidad de agentes ambientales, exposición de la población y salud pública.” (CB06/02/0024); Grupo de Investigación consolidado “Farmacología y Toxicología.” (2009/SGR/214) y Grupo de Investigación consolidado “Farmacología y Toxicología.” (2005/SGR/00826).
