Bacteriocinas de Lactococcus lactis aislados de quesos asturianos: Nisina Z y Lactococina 972

Abstract

Se ha analizado la producción de bacteriocinas en cepas de lactococcus aisladas de quesos artesanales asturianos, de este modo se han identificado productores de Nisina Z que producen la bacteriocina en leche y, además, presentan aptitudes tecnológicas adecuadas para su inclusión en un cultivo iniciador. Por otro lado, se ha descrito una nueva bacteriocina Lactococina 972, cuyo espectro de actuación se restringe al género lactococcus. Esta bacteriocina se ha purificado y se ha obtenido su extremo amino terminal. No se detecto ninguna homología con otros péptidos en las bases de datos. Su forma activa es un homodimero, cuyos monomeros carecen de actividad inhibitoria. El estudio de su modo de acción ha revelado que no se trata de un péptido permeabilizador de membranas. Su blanco de actuación está relacionado con la síntesis de pared y, en concreto, inhibe la formación de septos de división en las células sensibles. El determinante genético de la Lactococina 972 se encuentra en un plásmido de 11 KPB (PBL1). La secuencia del gen estructural ha revelado que esta bacteriocina se sintetiza como una prebacteriocina con un péptido señal típico. Este gen presenta un promotor funcional y se encuentra acompañado de otros, probablemente implicandose la inmunidad y/o regulación.
