Alteraciones de los Astrocitos en el envejecimiento y en la enfermedad del Alzheimer

Abstract

El tema de esta tesis ha sido el estudio de los mecanismos de neurodegeneración asociados al envejecimiento cerebral y el papel que desempeñan los astrocitos como neuroprotectores utilizando modelos animales in vitro e in vivo. Los astrocitos son las células más abundantes del sistema nervioso central y su función es muy importante, además de dar soporte trófico a las neuronas tienen una función anticitotóxica y modulan la actividad neuronal. El papel de los astrocitos y su activación en el envejecimiento es un proceso poco conocido. Un mayor conocimiento de esta célula glial puede ayudarnos a entender los cambios fisiopatológicos que conlleva el paso de los años en el cerebro humano. Hemos estudiado los efectos de Aβ42 sobre ls cultivos de los astrocitos humanos en la presencia o ausencia de los agentes prooxidantes como butionina sulfoximina (BSO), un inhibidor de síntesis glutatión y FeSO4 que libera hierro redox activo. Condiciones prooxidantes potenciaron la toxicidad del Aβ, que se demostraron por la generación de radicales libres, cambios inflamatorios y apoptosis en los astrocitos humanos. Un tratamiento similar fue ensayado en la rata in vivo. Una mezcla del Aβ40 y Aβ42 o sólo del Aβ42 fue infundida intracerebroventricularmente durante 4 semanas. Otros grupos animales eran simultáneamente infundidos con BSO y Fe. Un análisis realizado 4 meses más tarde mostró un mayor deterioro cognitivo en el laberinto acuático de Morris con el tratamiento de Aβ conjuntamente con los tratamientos con agentes prooxidantes. Agentes prooxidantes también potenciaron la patología del tejido cerebral, así los estudios histológicos mostraron una mayor reactividad astroglial, depósitos de Aβ y daño oxidativo en las neuronas hipocámpicas sensibles a EA.

Este trabajo ha sido realizado dentro del proyecto del Instituto de Salud Carlos III (PI-030467) titulado "Alteraciones de la función neuroprotectora de los astrocitos activados en el envejecimiento y la enfermedad de Alzheimer" y del proyecto del Ministerio de Educación y Ciencia (SAF2006-13092-C02-02) titulado "Mecanismos de envejecimiento cerebral: papel de GSK3β/CDK5 y de las sirtuinas en cultivos de neuronas y astrocitos". Silvia García Matas ha recibido una beca predoctoral de Formació de Personal Investigador de la Generalitat de Catalunya (FI) entre los años 2005 y 2008.
