El tratamiento con fluoxetina induce neurogénesis mediante la activación selectiva de la vía WNT/ß-catenina en células progenitoras adultas del hipocampo

Abstract

Se ha descrito que en modelos animales los antidepresivos inhibidores de la recampación de monoaminas incrementan la tasa de neurogénesis durante la etapa adulta, habiéndose propuesto que la generación de nuevas neuronas en el giro dentado del hipocampo es necesaria para el efecto comportamental de estos. Por otro lado, la vía WNT-BETA CATENINA desempeña un papel muy importante en el desarrollo del SNC, promoviendo la proliferación y diferenciación de las nuevas neuronas. Además, se ha demostrado que también interviene en los procesos de neurogénesis durante la edad adulta, regulando la progresión de ciclo celular. En este estudio se ha administrado el ISRS fluoxetina (10mg/kg/dia s.c.) a ratas macho Sprague-Dawley durante 14 días. Se emplearon técnicas inmunohistoquímicas para localizar BETA-Catenina con marcadores de diferentes estadíos del desarrollo neuronal, como son GFAP,SOX2, DCX, NeuN y BLBP. se clasificaron las células de la zona subgranular del GD en células madr4e quiescentes (GFAP +, SOX2 +), células progenitoras amplificantes ((GFAP - , SOX2 +), neuroblastos (DCX +), neuronas inmaduras (DCX + NeuN +) y neuronas granulares maduras (DCX + NeuN +) y se cuantificó el numero de células que expresaban BETA-Catenina para cada fenotipo celular. El tratamiento crónico con fluovetina incrementó selectivamente el número de células progenitoras amplificantes (31+- 3%) con acúmulos de BETA-Catenina mientras que en el resto de fenotipos no se observaron cambios estadísticamente significativos. Estos resultados sugieren que la fuoxetina activa selectivamente la vía de la Wnt/BETA-catenina en células progenitoras amplificantes promoviendo su salida del ciclo celular y su diferenciación para generar nuevas neuronas en el giro dentado del hipocampo.
