Generación de clorhidrinas de ácidos grasos en la pancreatitis aguda

Abstract

Antecedentes: La pancreatitis aguda severa causa inflamación y necrosis del tejido adiposo. En este estudio hemos evaluado la generación de ácidos grasos clorados en el tejido adiposo necrótico. Métodos: En un modelo de pancreatitis por taurocolato en ratas se analizaron los niveles de las clorhidrinas de ácidos grasos en tejido adiposo, ascitis y plasma. También se administraron clorhidrinas en la cavidad peritoneal y se obtuvieron macrófagos peritoneales para evaluar su activación. Finalmente se determinó el nivel de clorhidrinas en plasma de pacientes con pancreatitis aguda. Resultados: Se detectaron clorhidrinas de los ácidos oleico y linoleico en tejido adiposo y ascitis. En plasma se detectó la clorhidrina del ácido oleico. La administración de clorhidrinas indujo la expresión de TNFa en macrófagos peritoneales. En pacientes con pancreatitis severa los niveles de clorhidrinas fueron significativamente más elevados que en las leves. Conclusión: Durante la pancreatitis aguda se generan ácidos grasos clorados que pueden intensificar la respuesta inflamatoria. En pacientes, los niveles de clorhidrina permiten diferenciar inicialmente pancreatitis leves de las severas. .
