Procedimiento para determinar la activación del receptor de citocinas mediante el uso de un anticuerpo

Abstract

La presente invención se refiere a un procedimiento para determinar la activación del receptor de citoquinas, especialmente del receptor de la hormona de crecimiento (GHR), por medio del uso de un anticuerpo capaz de discriminar entre una conformación activada e inactiva del receptor de citoquinas, especialmente la conformacion de GHR o de la proteína de unión a la hormona de crecimiento (GHBP), comprendiendo el procedimiento: poner en contacto el anticuerpo y el receptor de citoquinas para formar un complejo, poner en contacto un ligando candidato con el complejo, medir la unión del anticuerpo al receptor de citoquinas inactivo y de esta manera discriminar entre una conformación activada y una conformación inactiva. También se refiere a un procedimiento para seleccionar agonistas y antagonistas funcionales de citoquinas, especialmente agonistas funcionales de la hormona de crecimiento (GH), entre ligandos candidatos, por medio del uso de un anticuerpo específico para la conformación del receptor de citoquinas activada. La invención también se refiere a un anticuerpo dirigido a los subdominios I y II que abarcan la bisagra de la región extracelular de la hormona de crecimiento y que es capaz de discriminar entre una conformación del receptor activada e inactiva. La invención también se refiere al uso del anticuerpo, por ejemplo, para la caracterización de interacciones del receptor de la hormona de crecimiento y para la selección de compuestos que estabilizan la dimerización de GHR.
