Transformación genética de Castanea sativa Mill. Y crioconservación de las líneas embriogénicas transformadas

Abstract

En el presente trabajo se ha estudiado el efecto del tipo de explanto y del genotipo en la transformación genética de líneas embriogénicas del castaño europeo (Castanea sativa Mill), así como su crioconservación. Los embriones somáticos aislados en el estado globular o grupos de 2-3 embriones en el estado globular o torpedo fueron los más efectivos para la transformación. Se logró la transformación en las siete líneas embriogénicas evaluadas, si bien el porcentaje de éxito es claramente dependiente del genotipo. Las líneas transgénicas fueron posteriormente crioconservadas utilizando el método de vitrificación. Así mismo, se han iniciado los ensayos para la transformación de estas líneas utilizando un gen (CsTL-1) que codifica una proteína tipo osmotina (taumatina), de carácter antifúngico que podría inducir tolerancia a la enfermedad de la tinta.
