Myxomatosis in the Iberian hares: epidemiology, risk factors and genetic basis of host susceptibility

Abstract

[EN] The myxoma virus (MYXV) is a poxvirus affecting lagomorphs with a remarkable evolutionary history. Its natural host is the tapeti (Sylvilagus brasiliensis), a South American species in which infection is generally innocuous. In the 20th century, MYXV was introduced into European rabbits (Oryctolagus cuniculus) in Europe and Australia to control overpopulation. In this species, MYXV produces a fatal disease, myxomatosis. Following the first introductions, myxomatosis caused mortality rates near 100%, drastically reducing rabbit numbers. However, attenuated viral strains and genetically resistant rabbits soon emerged. Consequently, disease impact decreased, and populations started to recover, though never to previous dimensions. Today, myxomatosis still influences rabbit population dynamics in their native range, the Iberian Peninsula, though disease severity is generally lower. In 2018, MYXV jumped to another lagomorph, the Iberian hare (Lepus granatensis), after the emergence of a recombinant strain, ha-MYXV/MYXV-Tol. The ha-MYXV includes a 2.8 kb insert, but otherwise shows high identity with strain previously circulating in rabbits (Lausanne, Lau-MYXV). Since 2018, outbreaks have been reported throughout the Iberian hare’s range. Yet, epidemiological studies have been challenging due to limited monitoring, largely restricted to passive surveillance. In this context, this thesis aimed to increase the understanding of myxomatosis in hares by combining various surveillance and diagnostic tools, as well as different statistical and genomic approaches.
