Abstract

Antecedentes y objetivo: Existe controversia para identificar qué metabolito final ejerce actividad biológica tras el consumo de polifenoles. El ácido elágico (nueces, fresas, granada, etc.) es metabolizado por la microbiota intestinal hasta urolitinas, metabolitos anti-inflamatorios, antiproliferativos, neuroprotectores, moduladores de la microbiota intestinal y protectores de la barrera intestinal. Urolitina-A (Uro-A) induce autofagia en modelos murinos de enfermedades del sistema nervioso central (SNC). Sin embargo, se desconoce si llega al parénquima cerebral para ejercer estos efectos. Nuestro objetivo es explorar los efectos de Uro-A en un modelo de ratón de envejecimiento fisiológico y comprobar si Uro-A y/o sus metabolitos de fase-II llegan al cerebro. Métodos y resultados: Administramos 2,3 mg de Uro-A i.p. diariamente (~14 mg/persona de 70 kg) en ratones jóvenes (6 m) y viejos (22 m) durante 8 semanas. Realizamos electrorretinogramas, pruebas de comportamiento, histología de retina, análisis de mitofagia y del metabolismo de Uro-A (UPLC-QTOF-MS). Uro-A mejoró el índice neurodegenerativo, memoria visual, daño oxidativo, integridad sináptica, gliosis, visión nocturna y mitofagia, especialmente en ratones viejos. Un estudio cinético mostró pico de Uro-A, Uro-A-sulfato y Uro-A-glucurónido en plasma, y de forma relevante, Uro-A y Uro-A-sulfato, pero no Uro-A-glucurónido en cerebro perfundido, 30 minutos tras administrar Uro-A. Conclusiones: Uro-A ejerce neuroprotección previniendo deterioro del SNC asociado al envejecimiento. La llegada al cerebro de Uro-A y Uro-A-sulfato tras administrar Uro-A vía i.p., sugiere que cruzan la barrera hematoencefálica. Uro-A-sulfato podría ser un reservorio para liberar Uro-A tras desconjugación por arilsulfatasas cerebrales. Sin embargo, Uro-A-glucurónido, relevante en humanos, no llega al cerebro.
