Producción de bioplástico a partir de PET con la bacteria Comamonas testosteroni

Abstract

El tereftalato de polietileno (PET) es uno de los plásticos más empleados del mundo, con el agravante de que, en la mayoría de las ocasiones, es de un solo uso, por lo que supone un grave problema medioambiental. Convencionalmente, el PET se recicla mediante técnicas fisicoquímicas que reducen su vida útil, generando con el tiempo un residuo inservible. Ante este problema,las aproximaciones biotecnológicas, basadas en el empleo de microorganismos y sus enzimas,surgen como una alternativa más sostenible. Se han descrito diversas enzimas microbianas capaces de despolimerizar el PET hasta sus monómeros fundamentales, siendo paradigmáticas la PETasa y la MHETasa de la bacteria Ideonella sakaiensis. La PETasa despolimeriza las cadenas de PET, dando lugar a Mono(2-hidroxietil)-tereftalato (MHET), el cual es hidrolizado por la MHETasa,quedando fi nalmente TPA y EG. La bacteria Comamonas testosteroni es capaz de crecer utilizando el TPA como única fuente de carbono y energía. Además, tiene la capacidad de formar polihidroxibutirato (PHB) a partir del TPA, un biopolímero muy apreciado en la industria por sus propiedades similares al polipropileno,pero con la ventaja de ser biodegradable. En nuestro grupo hemos empleado a Comamonas testosteroni como chasis sintético para la expresión heteróloga de una PETasa y una MHETasa recombinantes, de forma que la bacteria puede despolimerizar el PET y crecer a partir de sus monómeros, además de producir PHB. De este modo, planteamos una estrategia de economía circular para un reciclado alternativo del PET.
