El factor de splicing PRPF40B regula el crecimiento y la diferenciación neuronal.

Abstract

Introducción/Objetivos: PRPF40B es un homólogo humano del factor esencial de splicing Prp40 de levaduras. Nuestro grupo ha mostrado que interacciona con factores clave del spliceosoma y que modula eventos de splicing alternativo1, 2. PRPF40B ha sido implicado en la patogénesis de enfermedades neurodegenerativas y, en relación a ello, hemos mostrado que la eliminación del homólogo de Prp40 en Drosophila provoca alteraciones en la morfología sináptica, disfunción motora y degeneración neuronal. El objetivo de este estudio es conocer el papel funcional de PRPF40B en el sistema nervioso y dilucidar los mecanismos moleculares por los cuales regula la expresión génica neuronal. Metodología: Se han realizado estudios de proliferación, migración y diferenciación neuronal en líneas de neuroblastoma humano SH-SY5Y silenciadas de PRPF40B mediante CRISPR/Cas9. Asimismo, se ha llevado a cabo un estudio transcriptómico con el fin de conocer los genes diferencialmente expresados tras el silenciamiento de PRPF40B, hallando g
