Efecto del consumo de xilitol sobre el microbioma intestinal infantil

Abstract

Los edulcorantes son aditivos alimentarios empleados para sustituir el azúcar en la producción de alimentos y bebidas. Las autoridades de seguridad alimentaria advierten de sus potenciales efectos negativos sobre la salud, como incrementar el riesgo de diabetes tipo 2, entre otros. El xilitol es un edulcorantes ampliamente empleado, carente de un nivel máximo permitido. Ha sido estudiado en profundidad en cuanto a la salud buco-dental se refiere, sin embargo, los posibles efectos sobre la microbiota intestinal apenas han sido estudiados. Por ello, el objetivo de este trabajo ha sido evaluar el efecto dosis-dependiente del xilitol sobre el microbioma intestinal y su impacto sobre la función de barrera intestinal. El xilitol se administró a concentraciones crecientes al Simulador intestinal dinámico BFBL, estabilizado con microbiota fecal infantil. La composición taxonómica de la microbiota se analizó mediante secuenciación de amplicones de la región V3-V4 del gen 16S rDNA en la plataforma Illumina, y la formación de AGCC, mediante HPLC. Posteriormente, los sobrenadantes procedentes del simulador BFBL, se incubaron con la línea celular Caco-2 (modelo epitelial intestinal in vitro) para evaluar viabilidad celular e integridad de la monocapa celular. La alimentación del Simulador BFBL con xilitol a dosis crecientes, dio lugar a un incremento significativo de la abundancia relativa de la familia Lachnospiraceae y los géneros Blautia y Subdoligranulum de manera dosis-dependiente, acompañado de un aumento significativo en la formación de ácido butírico. Los sobrenadantes obtenidos durante la alimentación con xilitol no mostraron ningún impacto sobre la viabilidad celular ni sobre la integridad epitelial. Los resultados sugieren que la microbiota intestinal es capaz de fermentar el xilitol, favoreciendo el desarrollo de grupos microbianos productores de butirato en el ecosistema intestinal. No obstante, se requieren más ensayos para determinar su posible efecto fisiológico y el impacto que estos cambios tendrían sobre la salud del individuo.
