Abstract

La enfermedad de Alzheimer (EA) es un complejo trastorno neurodegenerativo que provoca un deterioro de la función cognitiva y la salud mental. Evidencias recientes sitúan a la microbiota intestinal como un importante factor de susceptibilidad en la EA al mostrar alteraciones específicas en la composición del microbioma de pacientes con Alzheimer y en modelos de roedores. Sin embargo, se desconoce si las alteraciones de la microbiota y su modulación por la dieta o factores ambientales pueden tener un efecto en el riesgo o la progresión a estadios más graves de la enfermedad. Para entender la implicación de los polifenoles de la dieta y el microbioma intestinal en la fisiopatología y síntomas de la EA, hemos llevado a cabo una intervención dietética durante meses en un modelo murino de taupatía (ratones transgénicos P301S) así como en ratones silvestres (C57 BL6/J) (N = 240 animales/genotipo) y se han evaluado factores neuronales, sistémicos e intestinales en el Alzheimer. Al final de la intervención con polifenoles, los resultados muestran una clara tendencia a la reducción de la activación microglial para el marcador Iba-1 en el hipocampo de los ratones y, por tanto de la neuroinflamación, altamente significativo en el modelo murino de taupatía. Durante el desarrollo de la enfermedad también se observaron cambios en el metaboloma fecal relacionados con la intervención dietética, dependientes del genotipo, y una disminución significativa de la abundancia relativa de distintos géneros y especies de la microbiota asociadas con deterioro cognitivo y demencia. Estos resultados indican que los polifenoles de la dieta podrian actuar en un proceso convergente mediado por el microbioma intestinal y los cambios neuronales derivados de la patogénesis de la EA, y por tanto favorecer mecanismos preventivos y de ralentización de la enfermedad, aliviando los síntomas y mejorando la calidad de vida de los pacientes.
