Sviluppo di nuove tecnologie in vitro per il miglioramento genetico del castagno e il rilancio della castanicoltura

Abstract

La tecnologia CRISPR/Cas9 rappresenta un ottimo strumento per effettuare modifiche target all¿interno del genoma, consentendo la disattivazione del gene di interesse. In questo lavoro è stata testata tale tecnologia su embrioni somatici di castagno, silenziando il gene della fitoene desaturasi la cui mutazione dà origine ad un fenotipo albino. I risultati hanno mostrato una buona efficienza di editing, di circa il 60%. Il costrutto CRISPR/Cas9 è stato anche testato sotto forma di ribonucleoproteina in protoplasti isolati da calli embriogenici. Sono stati isolati 4.500.000 protoplasti/mL (91% vitali) e trasfettati con il complesso ribonucleoproteico contenente l¿RNA guida (gRNA) disegnato sul gene della fitoene. I risultati hanno mostrato un¿efficienza di editing del 15-20%, paragonabile ai dati bibliografici
