Comportament fàsic de tensioactius derivats de glicerina: formació i estabilització d' hexosomes

Abstract

Els tensioactius no iònics derivats de glicerina son d’un gran interès en aplicacions industrials degut a la seva biocompatibilitat i biodegradabilitat en comparació a altres tensioactius no iònics convencionals.1 No obstant, el coneixement de les propietats físico-químiques d’aquests tensioactius es escàs. En aquest treball s’ha investigat el comportament fàsic del tensioactiu de grau tècnic monoisoestearat de digliceril en medi aquós, en funció de la temperatura i de la concentració. Les fases en equilibri s’han caracteritzat mitjançant observació visual amb llum polaritzada, microscòpia òptica i dispersió de raigs X a angle petit (SAXS). Els resultats han posat de manifest l’existència d’una fase de cristall líquid amb estructura hexagonal inversa (H2) en equilibri amb aigua, des de baixes concentracions de tensioactius fins al 96 %. El fet de no aparèixer una regió monofàsica, en contradicció amb la regla de les fases de Gibbs, es indicatiu de la presencia de diferents components en el tensioactiu estudiat. La presència de di- i triisoestearat de digliceril es va posar de manifest mitjançant tècniques d’espectrometria de masses (MALDI). La mida, polidispersitat i estabilitat de les dispersions de la fase H2 en aigua a baixes concentracions de tensioactiu (hexosomes) es van determinar per tècniques de dispersió dinàmica de llum (DLS) i retrodifusió de llum. Les dispersions estudiades van mostrar una baixa estabilitat que va millorar incorporant un copolímer de tipus bloc. La mida dels hexosomes (200 nm) va disminuir y es va poder controlar amb la concentració de polímer. El mecanisme d’estabilització es va investigar per SAXS. Els resultats han permès concloure que es produeix una forta adsorció de l’estabilitzant a la interfície aigua/hexosoma. Per altra banda, es va encapsular un principi actiu, la Ciclosporina A amb una activitat anti-immunogènica, en els hexosomes, aconseguint una eficàcia d’encapsulació elevada i un alliberament sensible al pH.
