Regulación de la citoquinesis por Rgf1 y la ruta de integridad

Abstract

[ES]La citoquinesis divide una célula madre en dos células hijas al final de cada ciclo celular y se lleva a cabo gracias al cierre de un anillo contráctil de actomiosina (CAR) y la síntesis del septo. En la levadura de fisión Schizosaccharomyces pombe, la ruta de inicio de la septación (SIN) asegura la coordinación temporal entre el ensamblaje del CAR y la constricción con la entrada de la membrana y la síntesis del septo de división, pero aún quedan preguntas sobre la regulación del CAR en condiciones de estrés. En este trabajo mostramos que Rgf1, GEF de la GTPasa Rho1, retrasa la finalización de la citocinesis bajo condiciones de estrés en la pared celular. Medimos por microscopía en tiempo real, a nivel unicelular, la formación, maduración y constricción del CAR en células silvestres y rgf1Δ tratadas con calcofluor (blankophor, BP). El BP no interfirió con el tiempo de ensamblaje del CAR ni con la tasa de constricción de este, pero retrasó el inicio de la constricción más allá del tiempo habitual determinado por el avance del ciclo celular. El retraso de la citoquinesis también se suprimió en ausencia de Pmk1, la MAPK de la vía de integridad celular (CIP), lo que lleva a una septación prematura y a un fenotipo de células encadenadas en condiciones de estrés leve. El retraso de la citoquinesis inducido por el estrés de la pared celular se correlaciona con el mantenimiento de la actividad de la ruta SIN y depende de esta ruta de señalización para que se logre. Por lo tanto, bajo el estrés en el septo, la vía CIP y la ruta SIN regulan la contracción del CAR para asegurar que la citoquinesis se complete de forma efectiva.
