Abstract

Introducción/Objetivo. Al envejecer aparece un deterioro de la respuesta inmunitaria, denominado inmunosenescencia, que está asociado a un estrés oxidativo. Recientemente hemos observado que la frecuencia de micronúcleos en los linfocitos T, lo que es consecuencia del daño al ADN por ese estrés oxidativo, aumenta al envejecer, y se ha propuesto como marcador de la incorrecta proliferación de estas células, hecho típico de la inmunosenescencia. Hemos comprobado que la ingestión de Akkermansia muciniphila (AKK) por ratones viejos, mejora significativamente el funcionamiento de las células inmunitarias y su estado redox, pero se desconoce si puede afectar la formación de micronúcleos. Por ello, el objetivo del trabajo ha sido comprobar si la toma de AKK puede mejorar esa frecuencia de micronúcleos en los linfocitos T y la proliferación de estos en ratones viejos. Metodología. En ratones hembra de la cepa ICR-CD1 viejas (72±4 semanas), se administró de forma diaria e individualizada AKK (2x108 ufc/100 μl PBS) durante un mes. El grupo control de la misma edad recibió solo el PBS. Trascurrido dicho mes se obtuvieron las células peritoneales, se enriquecieron en linfocitos T, mediante un protocolo de adherencia a placa, y se cuantificaron los micronúcleos utilizando la prueba de “Cytokinesis- Block Micronucleus (CBMN) Assay” basada en la inhibición de la citocinesis durante la mitosis para dar lugar a células binucleadas, que pueden contener micronúcleos detectables con hematoxilina-eosina. En paralelo se llevó a cabo el estudio de la proliferación de los linfocitos en respuesta al mitógeno específico de células T, concanavalina A. Resultados. En los ratones que habían ingerido el probiótico AKK una menor frecuencia de células con micronúcleos (P0,05), en comparación con los controles. Conclusiones. La ingestión de AKK puede ser una excelente estrategia para controlar la inmunosenescencia y conseguir un envejecimiento saludable.
