Estudio de genes de autofagia y la susceptibilidad a tuberculosis en españoles

Abstract

[ES] Estudios recientes han demostrado que la susceptibilidad a tuberculosis se encuentra parcialmente influenciada por autofagia y los diferentes alelos polim?rficos en genes relacionados con la autofagia (ATG) tiene un papel determinante en la respuesta del hu?sped hacia la infecci?n por Mtb. Analizamos las muestras de DNA de 54 individuos con tuberculosis latente, 78 con enfermedad tuberculosa y 230 controles de origen espa?ol. Realizamos un an?lisis por genotipos de los SNP (single nucleotide polymorphisms) ATG2B rs3759601, ATG5 rs2245214, ATG10 rs1864183 and ATG16L1 rs2241880 mediante sondas TaqMan. Hemos observado que la distribuci?n de genotipos entre los individuos sanos y pacientes con tuberculosis ha arrojado diferencias significativas para los portadores del genotipo GG de ATG2B rs3759601, pues estos individuos podr?an tener un menor riesgo de desarrollar la enfermedad. Individuos portadores del alelo ATG10 rs1864183 T podr?an tener una menor susceptibilidad a la infecci?n, si bien, tendr?a un rol protector a la infecci?n tuberculosa en este grupo poblacional. Existen diferencias en la distribuci?n de genotipos y alelos de variantes gen?ticas de autofagia y estas diferencias podr?an afectar, al menos de forma parcial, el riesgo y progresi?n de la infecci?n tuberculosa. Nuestros hallazgos podr?an tener relevancia en la comprensi?n que tenemos del control gen?tico de la respuesta inmune contra micobacterias, y sobre la progresi?n de infecci?n a enfermedad activa.
