Estudi in vitro dels mecanismes d'accio sinaptics dels isomers de l'hexaclorociclohexano

Abstract

El hexaclorociclohexano (hch) es un hidrocarburo clorado del que se conocen 8 estereoisomeros, de los cuales la forma gamma (lindane) tiene propiedades insecticidas y se le han descrito efectos toxicos. En el presente estudio se han realizado incubaciones de secciones regionales de cerebro para determinar el efecto de estos compuestos sobre la transmision sinaptica y el metabolismo de los inositoles-fosfato. Los resultados obtenidos han puesto de manifiesto que el lindane estimula la liberacion de noradrenalina (na) inducida por despolarizacion in vitro, mientras que el delta-hch tiene efectos inhibitorios. Por su parte, los agonistas gabaergicos no son capaces de alterar los efectos de estos compuestos. Asimismo, los bloqueantes de los canales de ca no modifican el efecto de los isomeros del hch, mientras que la activacion de la proteina quinasa c como la inhibicion de la calmodulina tienen efectos aditivos con el lindane. Tambien se ha visto una posible implicacion de los mecanismos dependientes de voltaje que intervienen en el transporte presinaptico de cl. Finalmente, se ha puesto en evidencia un efecto inhibitorio del delta-hch sobre la hisrolisis de fosfoinositidos, sugiriendo un posible efecto sobre los mecanismos implicados en la plasticidad neuronal.
