Desafiando el metabolismo de Rhodospirillum rubrum con monóxido de carbono: crecimiento a altas concentraciones de un gas tóxico

Abstract

La bacteria púrpura no del azufre Rhodospirillum rubrum, ha sido ampliamente estudiada por su gran versatilidad metabólica, pudiendo consumir una amplia gama de sustratos como el gas de síntesis (syngas) y ácidos grasos volátiles (VGA), como el acetato. Además, su complejo metabolismo les permite sintetizar bioplásticos (PHB), derivados del licopeno y combustibles (hidrógeno). En el presente trabajo se estudia el crecimiento anaeróbico de esta bacteria en diferentes concentraciones de monóxido de carbono (CO, principal componente del syngas) con acetato como fuente secundaria de carbono, en presencia o ausencia de luz. Como resultado, se observó un crecimiento en luz óptimo entre presiones parciales de CO (ppCO)= 0,2-0,6 bar. Sin embargo, una vez que el CO se consumió completamente, las células entraron en lisis después de unos días. Este fenómeno no se observó a altas concentraciones de CO en las que el mismo no se consumió completamente (ppCOinicial > 0,8 bar). La duración de la fase de adaptación aumentó a mayor ppCO inicial, demostrando los efectos tóxicos del CO. Si bien el acetato se consumió completamente en luz, en oscuridad su concentración final fue inversamente proporcional al CO consumido, demostrando la dependencia de CO como fuente de energía en ausencia de luz y con fuentes de carbono cuyo nivel de reducción es similar al de la biomasa. Estos resultados sugieren que es posible el crecimiento de R. rubrum (y concomitantemente la producción de PHB o licopeno) a altas concentraciones de CO, aunque mejorar la adaptación inicial es crucial para acelerar el proceso.
