A captive breeding program for the endangered seahorse Hippocampus guttulatus has been recently developed to support the conservation of this species in Galician coasts. Population and family genetic analyses, based on non-invasive sampling of the wild broodstockfounded in captivity, have been carried out by means of microsatellite markers to avoid inbreeding and losses of genetic diversity in culture. The paternity analysis of the progenies obtained allowed the identification of parents involved in the matings, yielding information about the mating system in captivity for this seahorse species. The estimation of relatedness coefficients between pairs of offspring with known relatedness allowed the selection of the most suitable parentage estimator. This information is useful for the stablishment andmanagement of the broodstock and for designing groups of individuals with minimum kinship and maximum genetic diversity as possible
