Abstract

Los vectores recombinantes de la invención tienen un inserto con la secuencia codificante para el gen de C7L de la vaccinia o de un gen homólogo bajo control de un promotor que permita su expresión. Los vectores dichos protegen a células en quienes son expresaron del desarrollo del apoptosis. La reinserción del gen de C7L en el genoma de NYVAC invierte el fenotipo apoptotic del virus, sin perder sus propiedades de la atenuación. La protección contra apoptosis confirió por C7L hace los vectores de la invención adecuados para producir vacunas vivas, aumentando la tiempo de la expresión y la presentación de los antígenos adicionales codificados en su genoma, o para preparar las composiciones para la terapia génica del extender-término para el accionamiento en células dianas.

The recombinant vectors of the invention have an insert with the coding sequence for the C7L gene of vaccinia or of a homologous gene under the control of a promoter that permits its expression. Said vectors protect the cells in which they are expressed from the development of apoptosis. Reinsertion of the C7L gene into the NYVAC genome reverts the apoptotic phenotype of the virus, without losing its attenuation properties. The protection against apoptosis conferred by C7L makes the vectors of the invention suitable for producing live vaccines, by increasing the expression time and presentation of additional antigens encoded in its genome, or for preparing compositions for extended-term gene therapy for actuation in target cells.
