Identificación de amiloides tipo BAP en la microbiota intestinal y evaluación de su papel en el desarrollo de enfermedades neurodegenerativas

Abstract

Estudios recientes han reconocido que los amiloides bacterianos son elementos funcionales y estructurales de la matriz extracelular del biofilm de muy diversas bacterias. El biofilm formado por la microbiota del tracto intestinal es uno de los más abundantes del cuerpo humano y, como tal, produce una enorme cantidad de amiloides. En este escenario, tiene lugar una compleja interacción entre el sistema inmune del huésped y los amiloides bacterianos que, bajo condiciones específicas, pueden provocar patologías relacionadas con el mal plegamiento de determinadas proteínas. En esta investigación proponemos que la disbiosis en el amiloma (amiloides de la microbiota intestinal) causada por bacterias patobiontes (bacterias residentes de la microbiota con potencial patógeno) puede actuar como un factor desencadenante de enfermedades. Utilizando las proteínas BAP como modelo de amiloides funcionales y los trastornos neurodegenerativos relacionados con el mal plegamiento de proteínas como modelo de enfermedad, hemos estudiado la interrelación entre los amiloides bacterianos y enfermedad. Para ello, en primer lugar, se ha determinado la composición de amiloides tipo BAP del patobioma gastrointestinal. En segundo lugar, se ha analizado si los amiloides tipo BAP pueden iniciar o acelerar la polimerización de proteínas amiloides humanas utilizando modelos de agregación de a-sinucleína en neuronas motoras en cultivo y en el nematodo Caenorhabditis elegans
