Aripiprazole and olanzapine are atypical antipsychotics. Both drugs can induce metabolic and cardiovascular changes and adverse drug reactions. Pupillography is a non-invasive and costeffective method to determine autonomic nerve activity during antipsychotic treatment. The aims of the study were to optimize and validate a liquid chromatography-tandem mass spectrometry (LC-MS/MS) method to measure drug concentrations; to evaluate the effects of aripiprazole and olanzapine on pupillary light reflex; prolactin levels, lipid and glucose metabolism; hepatic, hematological, thyroid and renal performance; adverse events and safety and to assess the relationship between these parameters and pharmacogenetics. Genetic variants in cytochrome P450 (CYP), dopamine receptor (DRD2, DRD3), serotonin receptor (HTR2A, HTR2C) and ATPbinding cassette subfamily B (ABCB1) genes, among others, may be associated with the pharmacokinetics and pharmacodynamics of antipsychotic drugs.
