La podocalicina (Podxl) es un modulador funcional del endotelio vascular

Abstract

Podocalicina (Podxl) es una proteína de la familia del CD34 que se expresa de forma abundante en la superficie luminal de los podocitos glomerulares del riñón en los que se ha postulado juega un papel antiadherente. Sin embargo la sobreexpresión de Podxl como proteína de fusión con la proteína fluorescente verde (PodxlGFP) en células CHO aumenta la capacidad celular de adhesión y migración sobre ligandos inmovilizados. El análisis de la integración de PodxlGFP en complejos multiprotéicos mediante electroforesis ¿Blue-native-PAGE¿ de lisados de células CHO- PodxlGFP muestran la integración de la proteína en dos tipos de complejos con pesos moleculares de aproximadamente 400 y 800 KDa. Los complejos de menor movilidad electroforética fueron analizados por espectrometría de masas y mostraron contener proteínas del proteasoma. El uso de inhibidores de la actividad proteolítica del proteasoma confirmó que Podxl es procesada en el proteasoma. Experimentos de co-inmunoprecipitación mostraron la asociación de PodxlGFP con ezrina y NHERF, proteínas que se ha descrito que interaccionan con Podxl en podocitos de riñón, lo que indica que la vía de señalización es la misma independientemente de la capacidad adherente y antiadherente de Podxl. La interacción de estas proteínas con PODXL en células CHO fue analizada utilizando líneas celulares que expresan de forma estable proteína de fusión con GFP mutantes de Podxl con deleción de todo el dominio citosólico o del dominio DTHL en el extremo citosólico. Los resultados indican que el dominio DTHL no es el único punto de interacción entre estas proteínas.
