Abstract

En este trabajo se presentan los resultados de aplicar por primera vez las condiciones fotoautotróficas al cultivo in vitro de castaño, a fin de incrementar la calidad de la planta durante las etapas de multiplicación, enraizamiento y aclimatación, y favorecer su propagación a gran escala. Para la etapa de multiplicación, los experimentos realizados han permitido diseñar un protocolo eficaz de cultivo sin aporte de azúcares exógenos, que se utilizó con 4 genotipos de castaño resistentes a la tinta. Este protocolo consiste en disponer segmentos apicales y basales en biorreactores de 10 L, usando medio líquido y dados de lana de roca como soporte, y someterlos a alta intensidad lumínica y ventilación forzada con aire enriquecido en CO2. Para el enraizamiento se utilizaron 15 genotipos que fueron tratados con inmersión basal en ácido indol- 3-butírico, seguido de cultivo en biorreactores de 16 L sobre bloques de lana de roca en medio sin azúcar y con altos niveles de CO2. Los porcentajes de enraizamiento y aclimatación fueron elevados y permitieron aplicar este sistema con eficiencia a todos los genotipos estudiados.
